Persently I own a house with my brother and 3 sisters. If i have my wife sign a Interspousal transfer Grant Deed, And I if die what would happen to my portion of the property in question, Can my wife make claim on it, or will the property portion go to my children?

From signing of this "Interspousal Transfer Grant Deed" your wife is eligible for portion of property after your death.The portion of "Community Property" will be benefited to your wife.

Sunil is wrong in saying that your wife will be benefited by signing the interspousal transfer deed. If your wife signs the interspousal transfer deed, it will become your separate property. She will have no claims on that property. After your death, your heirs can file an affidavit of heirship and claim the property.
